Tether Buffer

• Begin with approx. 700 mLs of DI H₂O
• Weigh out and add ingredients with constant stirring:

• 1M K₂HPO₄ 6.2mL
• 1M KH₂PO₄ 3.8mL (final conc. of KPO₄: 10mM)
• 10mM EDTA 10mL (final conc. of EDTA: 0.1mM)
• 3.3M NaCl 20.3mL (final conc. of NaCl: 0.067M)
• 1mM L-methionine 1mL (final conc. of L-met: 1µM)
• Glycerol 5g

• Bring up to 1 L with DI H₂O
• Autoclave
