YNB 5

YNB (1x) Agarose

For 2x YNB:

• Begin with approx. 300 ml of Milli-Q H₂O
• Weigh out and add with constant stirring:

o Yeast Nitrogen Base (without amino acids) 6.7g

• Bring up to 400 mL with Milli-Q H₂O
• Weigh out 20g of Glucose (Dextrose) and add to 100 mL of DI H₂O
• Autoclave YNB and glucose separately
• Allow to cool in 50deg. water bath
• Add glucose to YNB

Melt 500mL 2x pure agarose (see protocol) in microwave and cool in 50deg. water bath

Combine 2x YNB above with 2x agarose, mix and pour plates or divide into 2x 500ml aliquots in 1L bottles for later use.
